Sometimes you can get a good scan that way without removing it. If the glass lays flat on the scanner glass, you shouldn't get a glare. If you do get a glare, try scanning from several angles so you can take what is good from the different angles. Make your backup, and then follow the instructions and hints for using Photoflo and water.
